Highlights:

• Developed primary and secondary screens to discover anti-Herpes Virus compounds 6 months under deadline. Screened over 7000 characterized chemicals and substances resulting in several potential lead compounds. • Developed and performed first “blitz” mode assay in which up to 5000 extracts were assayed per day in semi-automated process. This exponential increase in throughput allowed for greater selectivity of “hits” resulting in the isolation of more novel compounds in less time. • Modified and ran 3 assays simultaneously at semi-automated high-throughput 96-well format (the HIV protease screen, a screen against a phosphatase in the insulin pathway, and the COX2 screen with COX1 counterscreen). • Modified and ran first 384-well format assay at Merck which increased throughput by a factor of 4 and significantly reduced assay costs. This assay targeted the convertase for tumor-necrosis factor, which was about to come under patent restrictions. • Selected to participate in 10 person “Core” team to restructure the Natural Products Drug Discovery department. All aspects of the department were analyzed and modifications implemented. My main contribution was the insistence on a department wide database. • Created and maintained inventory and database for 250,000-sample collection. Designed, programmed, and implemented procedure to create 4 sets of dilutions in 3 days, a process which had previously taken 3 months. • Experienced in sandwich and Competitive ELISA primarily for drug discovery. Designed, developed, and ran several assays using antigens/antibodies to bind enzymes/substrates to plate/bead surfaces.
