PROFILE Virologist/molecular biologist with over 15 years post-doctoral/research scientist experience in multiple research projects using a wide variety of molecular biological and cell culture techniques. * Cell culture and growth of pathogenic organisms in culture. * Experience with nucleic acid isolation/amplification of virus isolates from tissues and infected cell culture. Experienced with procedures and personal protective equipment (PPE) use in level 3 biocontainment laboratory (BSL-3). * Animal models of infectivity in BSL-3 laboratory conditions. * Field research experience - trapping and processing rodents under field BSL-3 conditions and processing of tissues for viral genome purification/amplification. * Training entry level lab workers at BSL-2 and in-lab training of graduate student at BSL-3. Experienced with creating stable transgenic cell lines using the lentiviral-based CRISPR/cas9 genome editing technology. * PCR, RT-PCR, quantitative (real time) PCR, primer design, optimization of cycling parameters * DNA extraction (prokaryotic and eukaryotic), RNA purification (eukaryotic) from cells and tissues * Restriction analysis, cloning, subcloning, PCR-based mutagenesis, transfection of eukaryotic cells, transformation of prokaryotic cells, DNA sequencing, Southern blotting * Spectrophotometric quantification of nucleic acids and proteins * Protein expression in bacterial and mammalian cells * Immunoserological assays (ELISA), immunoprecipitation, immunopurification of trans-expressed proteins, flow analysis, IFA (immunofluorescent assays) Experienced with animal models and husbandry including rodents (mostly mice) and insects (mosquitoes), dissection and tissue preparation (for viral titer, RNA purification, etc), blood collection for IgG titer, titer of neutralizing antibody response * Proficient with common software programs such as Word, Excel, PowerPoint, also with some more specialized software such as DNAStar, Prizm (scientific graphing), EndNote, on-line BLAST search, etc. * Fully trained and experienced in Biosafety level 2 and 3 working conditions/practices
