SUMMARY OF SKILLS/KNOWLEDGE * Cognizant of medico-legal and national laboratory accreditation agency (i.e. College of American Pathologist [CAP], Joint Commission on Accreditation of Healthcare Organizations [JCAHO], etc.) requirements in performance of all laboratory duties. Certified Laboratory Specialist in Molecular Biology by the ASCP (American Society for Clinical Pathology). Technical Supervisor for the Keesler Molecular Genetics laboratory. Molecular CAP inspector. Knowledgeable on laboratory safety and required personal protective equipment (PPE) in handling patient specimens in accordance with current hospital Infection Control directives. * Well-versed on laboratory principles, concepts, and methodologies associated with high-complexity genetic analysis. Ability to modify current protocols and/or develop new testing procedures with evaluation of associated supplies, reagents and equipment. * Knowledgeable on multiple DNA instrumentation systems (listed below) including multi-tasking computers controlling these instruments and associated quality control (QC) procedure and protocols. Operates and maintains instrumentation including: Biorad Imaging system, Stratagene Video Imaging System, ABI Verti, 2720 and 9700 Thermocyclers, Eppendorf MasterChef Thermocyclers, QIACube Robotic Extractor, Beckman Biomek 2000 or 3000 Robot System, Applied Biosystems (ABI) Prism 310 Genetic Analyzer, ABI 3130XL Genetic Analyzer, MiSeq, NextSeq 500, HiSeq 2500, ABI 7500 Real Time, ABI Quant 6, PSQ 96 MA Pyrosequencer, Luminex 200, Nanodrop 8000 Spectrophotomer. ABI Software used for analysis includes Genemapper, Genescan, Genotyper, SequencingAnalysis. NGS Sequence alignment and variant calling algorithms using Illumina BSO applications. External sequencing analysis programs include Sequencher 5.1. * Ability to extract DNA from whole blood, direct cells, buccal swabs and tissue specimens. Extractions for RNA from Nasal Swabs/Nasal Washes. Performs Polymerase Chain Reactions (PCR), RT-PCR, NGS sequencing, Restriction Digestions, DNA Sanger sequencing and Fragment Analysis via capillary and PAGE, Southern Blot via chemiluminescence, Dot Blot via chemiluminescence, MLPA and BAC labelling for FISH analysis. Limited experience with CGH, both BAC and Affymetrix SNP arrays. * Molecular tests performed include Cystic Fibrosis Fragile X (PCR and Southern Blot analysis), Hereditary Hemochromatosis, Factor V Leiden, BRCA NGS and Sanger sequencing, Spinocerebellar Ataxia, Spinal Muscular Atrophy (SMA) types 7 and 8, Rett Disease Sequencing, Connexin 26 Sequencing, Huntington's Disease, Myotonic Dystrophy (PCR and Southern Blot), Friedreichs Ataxia (PCR and Southern Blot), CGH, Pharmacogenetics and Y-microdeletion testing. Tests performed require detection, analysis, identification and quantitation of genetic mutations. RT-PCR testing methodology for Swine Flu using RNA from extracted nasopharyngeal specimens. (*Revised test to meet cost requirement allowing laboratory to perform CF testing for Department of Defense for less than 1/3 cost.)
