Daily Duties at Buck Institute For Age Research & Ucsd:

• Investigate the protein biochemistry and function of secretory granules of neuroendocrine chromaffin cells utilizing proteomic approaches - purification of granules, gel electrophoresis, Western blots, microLC-MS/MS, and bioinformatics. • Determine specific cleavage sites by analyzing the primary sequences and potential proteases (prohormone convertases) responsible for the cleavages. Proteomic methods were utilized – purification of granules from adrenal glands, 2D gel electrophoresis, mass spectrometry (MALDI-TOF) & bioinformatics analysis, Edman sequencing & bioinformatics, immunoassays. • Write a postdoctoral research proposal based on the above work to determine the responsible proteases for the processing by biochemical, proteomics, and molecular/cell biological methods. • Investigate the mechanisms of Huntington’s disease (HD) by analyzing HD brain tissues for differential protein expression and elucidating the fragments of mutant huntingtin protein, primary causative agents of the disease. Proteomic approaches were utilized that included biochemical & immunochemical methods (Western blots, IP, IHC), mass spectrometry, differential expression analysis by spot quantitation & immunohistochemical staining and microscopy. • Synthesize new hypotheses drawing from the results of the above projects and from other results in the literature. • Manage and coordinate multiple projects, which involved collaborators from several institutions as the first author investigator of the projects working with the PI.
