LA (awarded 6/2012) o Managed experimental timeline, prepared and updated SOPs for company o Conducted weekly conference calls with CSO, CEO, scientist, business mgr., dir. of Bioinnovation center and techs o Acted as immediate supervisor and managed team of both Minivax and LSU-employed researchers o Scientific editor for presentations prepared by Bioinnovation Center Director for Annual Minivax Conference o Attended technical program at annual Minivax conference o In vitro Antibody Dependent Phagocytosis and phagocyte- independent killing Assay o Characterization of in vivo antibody pharmacokinetic, in vivo therapeutic efficacy and protection assessments • Natural Killer cell- CD4+ T lymphocyte cross- talk during Pneumocystis infection - o Intratracheal murine inoculations o Isolation of lymphocytes from murine lung/ spleen tissues using magnetic and fluorescently labeled cell sorting (ARIA) o Characterization lymphocyte subsets via multicolor flow cytometry analysis (LSRII) o Adoptive transfer of lymphocyte subsets o Analysis of infection burden by reverse transcription real- time PCR assays (RT-PCR) o Development of in vitro co-incubation assay for pathogen killing and immune response analysis o Chemokine/ Cytokine expression analysis via multiplex assays • Prime- boost vaccination with a novel IP-10 - ovalbumin fusion construct enhances antigen specific local immunity and serum IgG titers - Designed project. Presented at American Thoracic Society General Conference o Intellectual Property and competitive research analysis o Design of expression plasmids and vaccination strategy o DNA plasmid injection using muscle electroporation o Intranasal inoculation of Adenoviral vectors o Isolation/characterization of lymphocytes & dendritic cells from murine lungs, lymph nodes & spleens (multicolor FACS analysis) o Analysis of antigen specific response (Intracellular cytokine staining, ELISPOT) o Cytokine quantification (ELISA) • Pneumocystis jirovecii sequencing project- o Purification of P. jirovecii organisms from clinical samples o Isolation of fungal DNA for the purpose of sequencing genome o Created standard from sequenced HSP70 gene via cloning for DNA quantification o Next-Generation sequencing analysis • Immunotherapeutic effects of ELR-CXCR3 chemokines in experimental Pneumocystis infection - Tested treatments using lab SOPs o In vivo pharmacokinetic studies: animal handling, rodent drug administration and blood draw o Mucosal gene therapy utilizing adenoviral vectors • Microbicidal effector functions of ELR-CXCR3 chemokines - Conceived and designed project o Utilized Radial Immuno-diffusion assay (RID) to measure minimum inhibitory concentration (MIC)
