Daily Duties at Center For Molecular Medicine And Immunology:

 Designing multi-epitopic synthetic peptides from sequences of poxvirus antigens (e.g. WR, NYVAC, Lister, Copenhagen, Wyeth Dryvax, NYCBOH etc…) retrieved from public database like GenBank.  Recruiting smallpox vaccinated human donors after written informed consent, for blood collection, processing and storage (Liquid nitrogen).  Testing donor sera for detection of poxvirus-specific antibodies including neutralizing antibodies using ELISA and PRNT respectively and donor PBMC for reactivity to new peptide candidates by ELISPOT and CFSE-based cell assays, after HLA typing by SSP-PCR.  Selecting and chemically conjugating the most reactive (or immunogenic) peptide candidates to HLA-DR-targeting antibodies and evaluating the new conjugates in HLA-DR-matching transgenic mice for antibody and T cell responses by ELISA to detect serum specific antibodies, PRNT to assay serum neutralizing antibodies, flow cytometry CFSE-based cell assays to monitor spleen T cell responses.  Collecting and analysing all the data for report in internal meetings with the higher management before presentation in external conferences and submission in peer-reviewed journals and maintaining written and computerized records of data, and lab stock inventory.  Designing, developing, and optimising methods, including molecular biology techniques, immunoassays, flow cytometry cell-based assays, cell cultures and poxvirus propagation and titration.  Writing experimental plans, protocols, SOPs and manuscripts, proposing new project ideas for grant writing purposes and identifying emerging technologies of great relevance for the project and the research centre.  Supervising a laboratory staff and collaboratively working with colleagues, technical staff, the PI and other researchers.
