Daily Duties at Harvard School Of Public Health:

• Assisted lead researcher in research planning, development, experimentation, data collection, analysis, and interpretation. • Carried out genotyping of mice, bleeding, organ/tissue harvesting and immunizing by diverse routes (Intraperitoneal, Intravenous, Intramuscular, Intradermal and Subcutaneous) • Optimized and performed various molecular biology and protein biochemistry techniques such as DNA/RNA extraction/preparation, affinity and gel filtration chromatography, PCR and real time qPCR, Western and Southern blot analysis • Optimized various immunological protocols such as ELISA, MACS separation, Flow cytometry, intracytoplasmic cytokine staining and FACS sorting and used the optimized protocols to perform Colon explant culture, T-regulatory cell isolation, primary T cell culture, isolation and culture of mouse dendritic cells. • Prepared tissue specimens, including gross processing, fixation and sectioning of both paraffin and frozen sections. • Developed and optimized histological protocols and performed immunohistochemistry on frozen and paraffin embedded sections. • Analyzed microarray gene expression data using Ingenuity Pathway Analysis (IPA) and confirmed it by real time qPCR. • My contribution to the projects resulted in publication of four high impact, peer-reviewed journals such as Cell, Cancer Cell, Cell Host and Microbes and PNAS.

Extensive experience in in vivo mouse models, Proficient in various in vivo techniques including pathology, surgery, various injections (intraperitoneal, intravenous, intramuscular, intradermal and subcutaneous), In vitro models of intestinal epithelial: Primary cultures, organ cultures and immortalized cells), bacterial transformation and lentiviral and retroviral generation and infection., Assays for signal transduction, proliferation, apoptosis, DNA damage, cytokine/hormone secretion, wound healing, barrier function, Molecular biology and protein biochemistry techniques: DNA/RNA extraction/preparation, affinity and gel filtration chromatography, PCR and real time qPCR, Western and Southern blot analysis, Immunological techniques: ELISA, Flow cytometry, intracytoplasmic cytokine staining and FACS sorting. Colon explant culture, primary T cell culture, isolation and culture of mouse bone marrow derived macrophages and dendritic cells. , Histological techniques: including embedding, sectioning, FISH, immunohistochemistry and immunocytochemistry, Microscopy: Fluorescent and light microscopy: Bright field, fluorescence and confocal microscopy for live cell and fixed cell imaging to study cell morphology, dynamic location of proteins, Proficient in in vivo mouse models, including enemas, colonic barrier function analysis, colonoscopy, bone marrow transplant, generating crosses, genotyping, husbandry, Bioinforamtics: Microarray/RNAseq gene expression data analysis using Ingenuity Pathway Analysis (IPA), Statistics and data analysis, Excellent oral and written communication skills. Competently convey information to both technical and non-technical audiences. , Experimental design/ execution, data presentation/ publication, literature review, interdisciplinary collaboration, mentoring and supervision
