• Northern Blotting: Processed RNA from cells; processed gels and transferred the blots; radiolabelled w/ 32P-dCTP, scanned the blot using a Phosphoimager; analyzed the data; calculated percentage of mRNA expression. Processed polysome gradients via Northern and determined the molecular weight distribution of various RNA of interest. • PCR: Cloned out specific DNA fragments using designed sense and anti-sense primers. • Primer Design: Designed primers with correct melting temperature, percent of GC content, and an optimal length to achieve the optimum yield from parental DNA template. • Gel Shift Assay: Performed validation studies involving the binding of radiolabelled RNA to protein extracts to determine the specificity of certain RNA-protein interactions. • Sequencing: Sequenced specific DNA regions to check for any gene mutations before any further experimentation was performed by the target identification/development group. • Tissue Culture: Performed cell passaging. Treated cells with various compounds of interest and processed these cells for RNA studies. • Cloning: Molecular biology techniques including subcloning, ligation, and transformation. Knowledge in cloning vectors such as TOPO, TOP10F1’, pGL3, pRc/RSV, pBluescript.
