Translational Oncology: • Responsible for procurement of human patient samples from various cancer types. • Processed PBMCs and BMMCs from patients’ whole blood and bone marrow aspirates. • Assessed monoclonal antibody efficacy using ADCC assay (chromium-51) with human PBMCs and BMMCs. Maintained sample database for human patient samples. • NK cell isolation from human PBMCs to be used in ADCC assay. • NK receptors profiling for healthy and patient PBMCs. • Proficient in LSRII Fortessa and designing 9-color flow cytometry assays. • Used Quantigene to correlate mRNA expression to FACS binding data as well as protein expression by western blot. Virology: • Maintained up to 7 mammalian cell lines for anti-viral study. • Extracted quality DNA, RNA, and proteins from cell cultures and processed them in downstream applications (RT-PCR, western blot, ELISA, and PCR). • Used fluorescent microscopy to visualize positive labeled cells to determine virus titer. Bacteriology: • Developed, optimized and validated numerous cell-based and sandwich ELISA systems. • Characterized anti-bacterial monoclonal and polyclonal antibodies through both in vitro (Opsonophagocytosis assays, ELISA, FACS) and in vivo (sepsis and organ infection models) assays. • Generated anti-bacterial antibodies through mouse immunizations, spleen fusions and hybridoma subcloning. General: • Helped setting up a BSL-2 laboratory. • Managed BSL-2 laboratory operations. • Prepared study reports and conducted critical experiments for patent, scientific manuscripts and data packages. • Provided reliable and reproducible high quality results to meet project timelines. • Wrote SOPs and study reports. • Supervised student interns. • Trained research scientists, research associates, and collaborators on experimental assays. • Prepared presentations routinely and presented during group meetings. • Played a key role as member of Environmental Health and Safety committee to ensure a safe environment for lab members. • Served as member of SOP committee providing feedback on preparing SOPs.
