• Studied the effects of an inhibitor of glutamate release on cancer cell growth: o Utilized in-vitro and in-vivo analyses to elucidate and map the genetic toxicity mechanisms o Assessed effects on gene networks both alone and in combination with various chemotherapeutic compounds including radiation therapy as a means to additively or synergistically increase efficacious effects of the compound. o Evaluated the pharmacokinetic and pharmacodynamic activities of experimental receptor antagonists both in vitro and their absorption, distribution, metabolism and excretion via various routes of administration in vivo. • Assisted in the production of a prodrug allowing for a decreased dosage resulting in an increased therapeutic effect: o Screened various prodrugs for their efficacy and ability to decrease cancer cell proliferation in in vitro assays prior to in vivo testing. o Performed in vivo toxicity studies of cleared compounds to assess the activities of phase I and phase II enzymes. o Developed and executed in vivo pre-clinical assessment of prodrugs following toxicology studies in tumor bearing animals. o Monitored, collected, analyzed, and interpreted both in vitro and in vivo study results. • Using U1 Adaptor technology specifically targeted and suppressed the expression of the proto-oncogenes: o Designed and implemented in vivo assays to test the targeting efficiency of adaptor molecules using xenograft models of tumor progression targeting a subset of human cancer. o Monitored animals for signs of toxicity and administered treatments via tail vein infusion for the duration of the studies. o Collected, analyzed, and interpreted all in vivo results. o Analyzed the treatment groups for a reduction of tumor cell growth in in vivo models of tumor progression and monitored induction of targeted pathways leading to cytotoxic cellular responses and decreased modulation of effector molecules known to play a role in tumor cell proliferation. • Showed biologically targetable, optical nanoprobes can be imaged in vivo with (a) deeper penetration than that possible using conventional organic or quantum dot probes, and with (b) shortwave infrared (SWIR) emission in a range with lower tissue absorption for real-time tracking of disease states and their responsiveness to therapeutic interventions. o Developed an in vivo model to visibly detect tumors in a transgenic mouse line of human melanoma. o Performed quantitative biodistribution of i.p. administered and tail vein infused REANCs using ICP-MS to validate physiologic distribution and accumulation of particles to various organs captured by imaging system. o Evaluated several formulations of rare earth particles in in vivo mouse models of tumor progression and metastasis to determine the feasibility of using SWIR to detect clinical disease and pharmacovigilance risk assessment.
