Daily Duties at Louisiana Tech University:

Biology: ● Cell culturing and replating cryopreserved rat brain glioblastoma cancer cells. ● Immobilization of consecutive layers of L-glutamate oxidase and mono-layer of brain cells in microfluidic device. ● Performed immunohistochemical cell staining using Trypan blue, Calcein AM, DAPI, MTT Assay, Fluo 3 etc. ● Cell imaging using fluorescence microscopy. ● Patterning of Brain Cells in selective areas by immobilizing cells on glass substrate using poly-l-lysine. ● Studied L-glutamate dynamics, cell metabolism and calcium signaling in brain cancer cells during normal conditions and by addition of stimuli such as Potassium Chloride, ATP, Cystine and Ionomycin. ● Glucose oxidase/Glutamate oxidase was coated on glass using layer by layer nano self-assembly technique for developing glucose/glutamate sensor. ● Kinetics of Immobilized glucose oxidase and glutamate oxidase activity was done using fluorescence spectrophotometer. Fabrication: ● Optimization and fabrication of thin film antimony-bismuth thermopiles and microfluidic devices using lift-off photo-lithography process involving semiconductors and etching techniques. ● Working knowledge of techniques such as Chemical Vapor Deposition (CVD), Physical Vapor Deposition (PVD), and Electrodeposition while depositing heavy metals in fabricating thermopiles. ● Performed PVD of bismuth and antimony on microscope glass slide and liftoff is employed to form thermopile electrodes. ● Prepared a SU-8 mold on the etched Si wafer employing Photolithography techniques and Reactive Ion etching, PDMS casting is done and the casted PDMS is peeled out of the mold to fabricate microfluidic devices. ● Demonstrated the Oxygen plasma treatment for bonding PDMS and glass slide and obtained microfluidic device. ● Designed an optimized photo mask layout of the thermopile electrodes and micro channels using Auto-Cad, and finite element analysis (CoventorWare MemCFD module) & COMSOL was used to simulate to obtain the thermal characteristics and flow dynamics. ● Characterized highly sensitive thin film antimony-bismuth thermopiles and measured Seebeck coefficient (90% theoretical value) and heat power sensitivity (83% efficient). ● Fabricated and characterized microfluidic thermoelectric calorimeter to measure dynamic temperature changes of order 10-4°C.
