# Aug 2004 – June 2009, Wayne State University & Karmanos Cancer Institute, Detroit, MI * PhD Thesis (Advisor: Dr. Ahmad R Heydari): Role of Uracil DNA glycosylase in BER pathway under folatedeficiency • The exact mechanism by which folate deficiency induces carcinogenesis is yet unclear. Folate deficiency increases the uracil pool which is repaired by DNA glycosylase initiated base excision repair (BER). • Thus, we identified whether DNA Glyocsylase (the first enzyme of BER pathway) silencing displays a protective phenotype under folate deficiency. So to do a proof of concept (POC) study we knocked down UDG and created a haploinsufficiency, then studied its affect on overall BER pathway and DNA damage response in tumor cells. • Research article describing this research work is submitted * Project 1 (2005-2007): Impact of Folate deficiency on epigenetic alterations of DNA and its role in Carcinogenesis • Folate deficiency alters the methylation pool with in the cell under oxidative stress (2NP), we next analyzed whether it effects promoter methylation of various BER genes, such as DNA glycolsylases, promoter have CpG islands. Additionally, we also established this could alter the transcription or enzyme activity of DNA glycosylases • Research article describing this research work is submitted
