Daily Duties at University Of California, Los Angeles:
I received a National Research Service Award to study the structure-function relationship in IgA & develop technology to produce recombinant IgA and secretory IgA in a single mammalian cell. My daily task was to train myself and students that were helping me in the following tasks. I also interacted with Dr. Sherie Morrsion on a daily basis to troubleshoot problems. a) Producing genetic expression vectors to express antibodies and novel proteins by teaching them how to manipulate DNA fragments using the Software Gene Construction Kit, how to design primers, how to set up PCR, and how to clone PCR fragments into expression vectors. b) Express the antibodies and novel proteins in myeloma and 293 cells c) Grow transfectants in large cultures, and to purify antibodies and novel proteins from culture supernatants by affinity chromatography using Pharmacia FPLC. d) Characterizing proteins by SDS-PAGE, Western Blotting and ELISA. e) Characterize the proteins in vitro using cell lines and in mice. f) To write up new protocols. g) To write manuscripts to publish in scientific journals.
