Studied the neuronal role of the glycolytic enzyme enolase (ENO) in Drosophila melanogaster during central nervous system (CNS) degeneration Developed ENO null flies using transposable elements (p-element) •Cloned ENO cDNA (1.4kb) into pcDNA, pET, pGEM and pUAST vector •Generated ENO clones with RFP, HA and HIS tags •Cloned ENO genomic sequence (5.2kb) into pCasper vector •Generated ENO mutations using QuikChange mutagenesis kit •Engineered flies that overexpressed wild-type and mutant ENO using the GAL4/UAS system • Knocked down the expression of ENO using RNAi in flies •Used RFP-tagged ENO to examine its cellular localization in HEK and COS cells •Measured the enzymatic activity of ENO using fluorescence (FLUOstar Omega) •Mapped ENO chromosomal insertion using Xa balancer flies •Expressed and purified ENO from HEK 293, BL21 and S2 cells •Executed ENO cDNA and genomic rescue experiments •Performed immunoprecipitation (IP) experiments to examine ENO interacting partners from HEK cell lysates and fly brains •Assisted students in designing fly genetic crossing schemes •Assisted students in planning and troubleshooting complex experimental paradigms in basic science research
