Daily Duties at Purdue University:
Study protein expression (such as band 3 and ankyrin), purification (ion exchange, affinity, and hydrophobic chromatography) from human RBCs and bacteria for protein complex, binding, and crystallization studies. Interpret and characterize structure and function of the human erythrocyte membrane through the experimentation designed. Oversee and manage biochemical assays on purified wild-type and mutant proteins such as GST binding assays and isothermal titration calorimetry (ITC) assays. Carry out in-depth data definition of docking interface between erythrocyte band 3 and ankyrin protein to characterize contact residues between ankyrin and band 3. Perform competition assays, immunoassays on purified wild type and mutant proteins. Identified differences between band 3 complexes and released structural heterogeneity within band 3 subpopulations through diffusion study in intact erythrocytes. Ensure safe and quality handling of bacterial (microbiology) cultures, including culture development and maintenance, manipulation,and protein extraction. Optimize protein yield.
