Daily Duties at University Of Pennsylvania:

• Design, construction and characterization of novel attenuated and recombinant Listerial vaccine vectors expressing HIV-1/SIV, or SHIV genes: such as IPTG inducible system, ssrA proteolysis tagging system (enhancing protein degradation), chromosomal integration system (different integration sites). The gene expression and translation were examined in details at the molecular levels (plasmid copy number, mRNA level, intracellular protein level). • Vaccine vector optimization, production and QC assay development for primate studies. • Studying mucosal immune responses by mucosal route delivery of Listerial vaccines (intranasal, oral, intravaginal and intrarectal immunization). • Culture, purification and characterization of recombinant adenovirus expressing HIV or SIV genes. • Studying the safety and immunogenicity of these recombinant strains in mice, especially examining the systemic and mucosal immune responses to HIV-1 or SIV of recombinant vaccines in heterologous immunization strategy (DNA or RNA vaccines, virus-like particles, dendritic cells, Listeria, adenovirus 5 or 26, poxvirus vaccines); Analysis of the polyfunctionality (CD107a, TNF-alpha, IL-2, IFN-gamma secretion ability) and phenotypes of antigen-specific CD8 T cells in effector, effector memory and central memory. • In vivo depletion of CD4 and CD8 T cells with monoclonal antibodies; Extensive study of in vivo cytolysis activity using CFSE labeling assay in many tissues (vagina, MLN, ILN, liver, Lung, spleen); Good at the adoptive transfer experiments. • Development of novel nucleotide-modified pseudouridine-containing mRNA-based HIV vaccines (mRNA, mRNA-loaded dendritic cells), and their formulation; Test the immune responses with mRNA as vaccines in mice in prime/boost strategy (Listeria /mRNA, Listeria/ DC, mRNA/adenovirus). • Attend the oncology seminars to keep updated knowledge in cancer cell biology, biomarkers identification and target therapies.
