The guinea pig produces a very robust T cell response, like humans, against intracellular pathogens but lack available reagents to do sophisticated molecular immunological studies. There was a need for reagents to study the guinea pig model for cellular immune response in otitis media. Using regions of homologous sequences from other species, I did data base searches for nucleotide and amino acid sequences, designed primers based on similarities found among the published gene sequences in other species; optimized the PCR reactions in the amplification of candidate genes; performed nucleotide sequencing using an automated fluorescent DNA sequencer; and, verified unknown sequences using Bioinformatics analysis. The goal was to develop an animal model that mimics human pathobiology in infectious disease which is necessary in company's pre-clinical vaccine development. Next, perform early development work on vaccines to Haemophilus influenza, Moraxella catarrhalis, and Staphylococcus aureus to illicit an authentic antigenic response which included designing gene-specific PCR primers and PCR amplification of the genes; sequence analysis of all the PCR amplicons, in vitro translation of the encoded proteins; and, purification and re-folding back to the native conformation to ensure proper immune recognition.
