*develop and maintain Genetic Mouse models (in vivo model) in cluding ► care and breeding ►genotyping ► characterisation and utilize ► injection ► immunization ► dissection ► cell isolation *Cell culture analyses of ► mammalian cell lines and ► primary cells (including ► transfection ► stimulation ► cell assays) *analyses at the molecular level of knock out cells (including ► Plasmid purification ► DNA / RNA isolation ► cDNA conversion ► DNA / siRNA-Transfection ► TaqMan RT-PCR (TaqMan® real-time PCR/qRT-PCR) ► RT-PCR (Real-time PCR) ► PCR (polymerase chain reaction) ► Site-directed mutagenesis ► Sequencing ► * Protein Biochemistry (Biochemical/Cellular Assays): ► Handle immunohistochemistry (IHC) ► immunoprecipitation (IP) ► SDS-PAGE ► native gel ► Western blot ► protein over expression and purification (bacterial- and mammalian recombinant protein expression) ► biochemical characterization of proteins (phosphorylation, sumoylation) ► in vitro caspase cleavage assay ► colorimetric GTPase assay ► cytokine-assay/enzyme-linked immunosorbent assay (ELISA). * Molecular Biology: ► Obtain experience on plasmid purification ► DNA/RNA isolation ► cDNA conversion ► DNA/siRNA-Transfection ► TaqMan RT-PCR (TaqMan® real-time PCR/qRT-PCR) ► RT-PCR (Real-time PCR) ► PCR (polymerase chain reaction) ► site-directed mutagenesis ► sequencing ► CHIP-assay (Chromatin Immunoprecipitation) * Microscopy: ► Administer fluorescence/confocal microscopy. • teaching at the NIEHS Summer Internship Program 2011 (lecture series within the Summers of Discovery (SOD) program) • mentored undergraduate students; supervised and designed student research projects
