Daily Duties at Research Institute Of Health Science (Rihes) :
1.HIV-1 coreceptors CCR5 and CXCR4 play an important role in viral entry and pathogenesis. To better understand the role of viral tropism in HIV-1 transmission, I studied the coreceptor utilization of viral isolates obtained from women enrolled in a study of heterosexual transmission in northern Thailand. Briefly, PBMC from HIV infected blood is prepared then co-cultured with PHA stimulated PBMC (blast). Culture fluids are collected every the other day to check the level of virus by particle-associated reverse transcriptase (RT) assay. The isolated virus was frozen as well as subjected to receptor study. 10 of GHOST cells expressing receptors eg. CCR5, CXCR4 were plated and fed with isolated HIV virus. The wells were observed under microscope for evidence of syncytia induction. Viral growth was monitored by quantification of p24 antigen. In addition, I diagnosed the HIV-1 from the babies who were born to HIV-infected mother (who were enrolled in the maternal transmission project) by PCR 2.CRF01_AE and subtype B have dominated the HIV-1 epidemic in Thailand since 1989. Currently, a new circulating recombinant form of HIV-1, CRF15_01B, as well as other unique CRF01_AE/B recombinants has been reported among prevalent HIV infections in Thailand. The aim of in this project is to study molecular picture through the assessment of subtypes among HIV-1 infected drug users in northern part of Thailand. My responsibility in this project was to isolate PBMC from 39 HIV seropositive (determined by EIA and Western blot) by Ficoll gradient centrifugation then DNA was extracted using QIAmp blood extraction kits (QIAgen).
