Fairfield, NJ. 12/2007 – 12/2009 Research Scientist - Protein Chemistry/Process Development Department • Individually carried out research for Novel Amidated Peptides Research project (SBIR grant #5R44DK063812-03) • Performed and developed a MALDI-TOF coupled MALDI-TOF/TOF based proteomic approach for amidated peptide discovery • Cell culture of prostate cells (DU-145 & PC3), and rat pituitary cells (CA-77) • cDNA library production from cell lines of particular interest. • Developed siRNA transfection and knockdown procedures for peptidylglycine alpha-amidating monooxygenase in two different mammalian cell lines. • Engineered DNA probes for PCR purification and sub-clone library production for peptides of interest • Developed RP-HPLC-UV/VIS, fluorescent, and refractive index detection systems for enzyme activity assays. • NCBI Blast, Entrez, SignalP 3.0, ExPASy Proteomics Server Unigene Laboratories, Inc. Fairfield, NJ. 10/2005 – 12/2007 Post-doctoral Research Fellow - Protein Chemistry/Process Development Department • Developed and carried out quantitative fluorescent and luminescent enzyme based assays (to detect nanomolar concentrations of amidated peptides). • Engineered liquid chromatography enzyme purification protocols, utilizing column and TFF (tangential flow filtration) systems. • Designed a Mass Spectral procedure for the discovery of novel amidated hormones/peptides. • FPLC and HPLC process development for peptide purification (isolation) from conditioned cell media. • Carried out basic R & D on novel amidated peptide assays. • Wrote SOP’s for instrument use and batch records for developed purification protocols.
