Daily Duties at Memory Pharmaceuticals:
Conducted ADME-tox assays in GLP and non-GLP environment to determine the safety profile of new drug candidates for CNS development. Performed metabolic stability assessment assays on lead compounds utilizing human, mouse, rat, dog, and monkey liver microsomes. Ran metabolic stability assays using cryopresserved hepatocytes to look at both Phase I and Phase II metabolism and induction, and determination of compound clearance. Performed various HTS Cytochrome P450 inhibition assays employing homogenous fluorescence technology to determine IC50 values for drug candidates. These assays were used to screen for potential inhibitors of human catalytic activity. Performed Caco-2 assay on potential drug candidates to assess cellular permeability and elucidate mode of transport. Assisted the Biology team with R&D in CNS transporters, L-type Ca2+ channels, ligand gated ion channels, Phosphodiesterase and G-Protein Coupled Receptors for drug design and development. Set up screening of compounds using Ca2+ sensitive dye with FLIPR or IMAP fluorescent screening with Analyst or Luciferase luminescence for cell based assays. Assisted with performing PK studies in rats, including preparing formulations, dosing and bleeding of rats, as well as collecting urine and fecal samples for analysis.
