Daily Duties at University Of California, Berkeley:

• Use single-molecule optical trapping techniques to study energetic and dynamic behavior of the different levels of chromatin—array of 8-12 nucleosomes, dinucleosome, and mononucleosome. Designed DNA fibers containing 1, 2, or more nucleosome positioning sequences. Optimized the techniques for in vitro formation of the various nucleosomal fibers and for the single molecule studies—bead:nucleosomal fiber ratio, buffer conditions. Aided in the construction of the dual beam optical trap used for the studies • Use single-molecule optical trapping techniques to study the condensation of DNA by positively charged synthetic spherical polymers (dendrimers) of various sizes. Investigated whether or not these synthetic polymers could mimic chromatin compaction, as well as how size and charge of these spherical polymers can affect DNA compaction. Assessed optimal buffer conditions to minimize non-specific interactions between the highly charged dendrimers and the microchamber/bead surfaces and of an efficient delivery system for the dendrimers.
