Daily Duties at University Of California, San Francisco:
• Investigate various extracellular matrix fragments for their potential in promoting healing after myocardial infarction. Using human umbilical vein endothelial cells (HUVECs) along with standard cellular and molecular biology techniques, performed in vitro assays to determine the ability of the fragments to promote adhesion, proliferation, migration, and activation of proteins involved in the angiogenesis pathway. Isolated monoclonal antibodies from hybridomas and chemically conjugated the fragments to the antibodies in order to target the fragments to the injured site in a rat myocardial infarct model. Used echocardiography and immunohistochemical and immunofluorescence techniques to monitor the ability of the fragments to improve heart function and promote angiogenesis within the infarct zone. • Investigate conducting polymers in treating heart disease. Conducted in vitro studies to determine cell viability and proliferation on the polymer scaffolds using HUVECs. Determined the conductivity of the various polymers using a four-point probe technique. Used a rat myocardial infarct model to assess the ability of the conducting polymer scaffolds to promote angiogenesis and wound healing. Performed some electrophysiology studies to determine the response the treated and untreated rat hearts to electrical stimulation. • Investigate the effect of peptide modified biomaterials on cell viability and proliferation. Conducted in vitro studies using human mesenchymal stem cells, human endothelial progenitor cells, and HUVECs to assess cell survival, proliferation, and angiogenic potential on the various modified biomaterials. Isolated the human endothelial progenitor cells for donor blood. • Investigate the effect of various biomaterials, extracellular matrix fragments in stem cell differentiation into electrically conducting cardiomyocytes. Performed electrophysiology studies using optical mapping and microelectrode array techniques on mouse embryonic stem cells, human embryonic stem cells, and mouse cardiomyocytes.
